Born in New York City in 1944, Matthew Cowles is the son of actor and theater producer Chandler Cowles. In a profile of his wife in The New York Times, actress Christine Baranski, he is described as "the black sheep member of a family with ties" to Drexel banking and the Cowles Publishing Company.

In 1966 Cowles acted in the short-lived Edward Albee adaptation of James Purdy's comic novel "Malcolm" on Broadway. His first small screen role was Joe Czernak in the series "NYPD" in 1969.

Cowles created and wrote the role of Billy Clyde Tuggle himself. He is an avid motorcycle rider.
